Azorubine (Carmoisine, E122): Banned in the United States for food use. Synthetic azo dye derived from coal tar and petroleum-based chemicals (produced from aniline derivatives and benzene), consisting of two naphthalene subunits with the chemical formula C20H12N2Na2O7S2. Originally listed in the United States in 1939 as Ext. D&C Red No. 10 for use only in externally applied drugs and cosmetics but was delisted in 1963 when no party was interested in supporting the safety studies needed to establish safety and was never used in food in the United States. Currently banned for food use in the United States, Norway, Sweden, and Japan. Permitted in the European Union since 2008 with strict mandatory warning label requirement (effective 2010) stating "may have an adverse effect on activity and attention in children" based on the 2007 Southampton Food Standards Agency study. The independent Southampton study, conducted by researchers at the University of Southampton and described by the UK Food Standards Agency as "a scientific study of the highest quality," found significant association between mixtures containing Azorubine (along with five other synthetic dyes and sodium benzoate preservative) and increased hyperactivity in both 3-year-old and 8-9-year-old children in the general population in a randomized, double-blind, placebo-controlled trial published in The Lancet. Independent peer-reviewed animal studies documented multiple severe toxic effects: hepatocellular damage with significantly elevated serum ALT and AST enzyme levels indicating liver toxicity at both low and high doses (2010 study), kidney toxicity with elevated urea and creatinine, impaired antioxidant status with decreased glutathione (GSH), superoxide dismutase (SOD), and catalase levels, elevated malondialdehyde (MDA) indicating lipid peroxidation and oxidative stress, altered inflammatory biomarkers, and adverse effects on liver histology. A 2024 independent study published in Food and Chemical Toxicology using zebrafish larvae documented developmental toxicity including increased mortality, morphological deformities (reduced growth, pericardial edema, yolk sac edema), oxidative stress with impaired antioxidant enzyme responses, cellular and lipid damage, and apoptotic cells observed in the neural tube and notochord. Reproductive and fertility harm documented in independent animal studies showing significant down-regulation of fertility-related genes (Testin, GDNF, c-KIT, FSHR, PRKA3, Spata7, SSEA1), decreased sperm counts in dose-dependent manner (49.00±0.28 at high dose versus 147.00±3.2 in controls), and testicular histopathological damage at doses above the Acceptable Daily Intake of 4 mg/kg body weight. Can trigger severe allergic reactions including skin rashes, hives, eczema, and asthma attacks, particularly in individuals sensitive to salicylates or those with existing asthma, as Azorubine acts as a histamine releaser. May contain aluminum as an undisclosed contaminant when produced as aluminum lake colors, with aluminum suspected of promoting neurological conditions including dementia, Alzheimer's disease, Parkinson's disease, and attention disorders. The dye is metabolized by reduction in the intestinal bacteria and liver cells into aromatic amines which have documented carcinogenic properties. Independent in vitro studies showed the dye inhibited the oxidative activity of rat liver succinic oxidase enzyme by approximately 40 percent. Children are particularly vulnerable due to documented hyperactivity effects in controlled studies, with the UK Food Standards Agency recommending voluntary phase-out and the Hyperactive Children's Support Group recommending elimination from children's diets. While EFSA concluded no evidence of mutagenicity or carcinogenicity based on standard genotoxicity assays and maintained the ADI at 4 mg/kg body weight, recent independent studies (2024) documented chromosomal aberrations in zebrafish larvae and cell apoptosis at doses close to the ADI, leading researchers to conclude "despite the low intake as a food additive, Azorubine may pose a health problem, therefore its use must be carefully monitored."
